Die Biobank des CCCF enthält Proben von Tumoren, die von den operativ tätigen Mitgliedsabteilungen reseziert werden. Das asservierte Gewebe steht Arbeitsgruppen des CCCF im Rahmen der interdisziplinären Forschungskooperation zur Verfügung.

Nutzen der Biobank

Biobanken sind für den medizinischen Fortschritt von immenser Bedeutung. Die dort eingelagerten  Patientenproben ermöglichen zusammen mit den zugehörigen patientenspezifischen Daten eine moderne biomedizinische Forschung. Hieraus werden neue medizinische Erkenntnisse für die Diagnose und Therapie von Erkrankungen gewonnen. Damit nehmen Biobanken eine Brückenfunktion in der translationalen Forschung ein. Die Anliegen und Autonomie unserer Patienten/Patientinnen stehen bei uns stets im Vordergrund.

Die Biobank des Comprehensive Cancer Center Freiburg (CCCF) sammelt projektspezifisch und projektunabhängig Frischgewebeproben und deren Derivate von Patient*Innen, die am Tumorzentrum Freiburg behandelt und versorgt werden. Dem Gewebe zugehörige klinisch-pathologische Daten werden den entsprechenden Gewebeproben angegliedert und in der Biodatenbank gespeichert.

Die Biobank des CCCF besitzt eine interdisziplinäre Infrastruktur und ist am Universitätsklinikum Freiburg im Institut für Klinische Pathologie lokalisiert. Ein Qualitätsmanagementsystem mit harmonisierten SOPs (Standardvorgehensweisen) und präzisen Verfahrensanweisungen bietet die Gewissheit, dass verbindlich geregelte Arbeitsschritte auf höchstem Niveau durch jeden Mitarbeiter und jede Mitarbeiterin  ausgeführt werden. Die Biobank asserviert qualitativ hochwertige Patientenproben und die zugehörigen klinisch-pathologischen Daten. Die kontinuierliche Weiterentwicklung des Biobankings als unverzichtbare Ressource der Krebsforschung ist ein erklärtes Ziel des CCCF sowie des Instituts für Klinische Pathologie. Hierbei gilt es, sowohl wachsenden Datenschutzansprüchen als auch den Entwicklungen auf der technischen Seite gerecht zu werden.

Zum Zweck einer überregionalen Vernetzung von Biobanken ist es unverzichtbar, harmonisierte Konzepte zu entwerfen, um an verschiedenen Standorten gleiche Voraussetzungen zu schaffen. In diesem Sinne kooperiert die Biobank des Standorts Freiburg im Deutschen Konsortium Translationale Krebsforschung (DKTK) mit den klinischen Forschern der Forschungsprogramme und weiteren Standorten des Konsortiums.

Die Biobank des CCCF wird durch das Institut für Klinische Pathologie des Universitätsklinikums Freiburg betrieben und durch dessen Ärztlichen Direktor - Prof. Dr. Martin Werner geleitet. Die Biobank wurde im Jahr 2002 gegründet und ist seit 2008 an das CCCF angegliedert.

Zugriff auf Gewebeproben haben Forscher*Innen nach Ausfüllen eines Projektantrages und deren Bestätigung durch das CCCF Board Shared Resources.  Im Projektantrag stellt die Forscherin/der Forscher das Forschungsvorhaben dar, nennt Art und Zahl gewünschter Proben sowie ggf. gewünschte Serviceleistungen der CCCF-Biobank, die er in Anspruch nehmen möchte. Alle Forscher*innen verpflichten sich, die geltenden Datenschutzrichtlinien einzuhalten. Transparenz

Transparenz stellt eine wesentliche Grundlage des Biobankings dar. Sie erfordert die Dokumentation des Umgangs mit den sensiblen Proben und Daten unserer Patient*Innen, der wir schon gemäß den Anforderungen guter wissenschaftlicher Praxis nachkommen. Weitergehend ist Transparenz ein wichtiges, grundlegendes Instrument zum Schutze von Spenderinteressen, welche die Kontrolle der Sammlung, Speicherung und Weitergabe von Proben und Daten unabdingbar macht. Zusätzlich dient Transparenz den Interessen der Forscher*innen, da die Akzeptanz und Kooperationsbereitschaft von Spendern und Spenderinnen erhöht wird. Die Verwendung der Biobankproben kann zum Zeitpunkt der Entnahme nicht im Detail definiert werden, sondern gilt einem allgemeinem Forschungsnutzen, im Rahmen aller ethischen und rechtlichen Richtlinien. Aufgrund dieser allgemeinen Regelung haben Spender*innen jederzeit die Möglichkeit allgemeinen Sammlungs-, Speicherungs- und Weitergabemodalitäten nachzuvollziehen. Die Spender*innen können jederzeit und kurzfristig ihr Recht auf Widerruf der Nutzung ohne persönliche Nachteile geltend machen. Proben, welche bereits in Forschungsprojekte eingebunden sind, können aus logistischen Gründen nicht nachträglich entfernt werden. Die dauerhafte Sicherung von Transparenz bietet entsprechend der Empfehlungen des Deutschen Ethikrates die Gewährleistung für den Umgang mit projektunabhängigen Sammlungen und die zum Zeitpunkt der Einwilligung bestehende Unkenntnis über den Forschungszweck der Proben.

Gemäß den Empfehlungen des Deutschen Ethikrates finden Sie hier folgende Angaben:  

  1. Rechtsform, datenschutzrechtlich verantwortlichen Stellen, Datenschutzbeauftragten, zuständigen Aufsichtsbehörden 
  2. Zuständigkeiten im Organisationsbereich der Biobank  
  3. Ansprechpartner und Möglichkeiten zur Gewinnung vertiefender Informationen  
  4. Darstellung der Zwecke und Regeln des Sammelns sowie der Verwendung und Weitergabe von Proben und Daten  
  5. Berichte über die Aktivitäten der Biobank 
  6. Maßnahmen der Qualitätssicherung

Zu 1. Rechtsform, datenschutzrechtlich verantwortlichen Stellen, Datenschutzbeauftragten, zuständigen Aufsichtsbehörden.

Die Biobank des Comprehensive Cancer Center Freiburg wird durch das Institut für Klinische Pathologie des Universitätsklinikums Freiburg betrieben und durch dessen Ärztlichen Direktor - Prof. Dr. Martin Werner - geleitet.

Alle Mitarbeiter*innen der Biobank werden entsprechend §6 des Landesdatenschutzgesetzes belehrt, in dem es heißt: „Den bei öffentlichen Stellen beschäftigten Personen ist untersagt, personenbezogene Daten unbefugt zu verarbeiten oder sonst zu verwenden (Datengeheimnis). Das Datengeheimnis besteht auch nach Beendigung ihrer Tätigkeit fort.“  Daten und Gewebeproben werden zunächst bei Einlagerung/Speicherung pseudonymisiert und vor Ausgabe erneut pseudonymisiert, so dass eine Identifizierung der Patientin/des Patienten nach heute geltenden Standards nicht möglich ist. Ferner ist es allen Mitarbeiter*innen untersagt, pseudonymiserte/anonymiserte Daten zu „re“-identifizieren.

Zu 2.  Zuständigkeiten im Organisationsbereich der CCCF-Biobank  

Die CCCF-Biobank wird vom Ärztlichen Direktor des Instituts für Klinische Pathologie geleitet. Der CCCF-Biobank sind ein(e) ärztliche(r) Mitarbeiter*in, zwei technische Assistent*innen, eine Schreibkraft sowie Sachmittel zur Asservierung, Lagerung, Qualitätskontrolle und Ausgabe von Frischgewebsproben und davon abgeleiteten Derivaten durch das CCCF bzw. das Klinikum zur Verfügung gestellt. Die CCCF-Biobank nutzt räumliche und apparative Ressourcen des Instituts für Klinische Pathologie.

Die Biobank ist in das CCCF als Shared Ressource integriert und im Institut für Klinische Pathologie lokalisiert. Die Biobank hat einen vom Vorstand des CCCF benannten Leiter und vom CCCF zur Verfügung gestelltes Personal, das für die Asservierung, Lagerung, Qualitätssicherung und Ausgabe von Bioproben und deren Derivaten sowie die Verknüpfung mit klinisch-pathologischen Daten verantwortlich ist. Leitung und Personal der CCCF-Biobank sind dem Ärztlichen Direktor des Instituts für Klinische Pathologie unterstellt und in dessen Qualitätsmanagement eingebunden. Über die Nutzung des für wissenschaftliche Zwecke von den Patient*innen zur Verfügung gestellten Biomaterials und der davon abgeleiteten Derivate entscheidet das Board Shared Ressources. Das Board Shared Ressources des CCCFs setzt sich aus dem ärztlichen Direktor des CCCFs, dem Sprecher der operativen Disziplinen, sowie dem Leiter des Instituts für Klinische Pathologie zusammen.

Zu 4. Darstellung der Regeln und Zwecke über Sammlung, Verwendung und Weitergabe von Proben und Daten

Die CCCF-Biobank dient der Förderung medizinischer Forschung. In der Biobank werden Gewebeproben und zugehörige Daten langfristig aufbewahrt und ausschließlich für die medizinische Forschung zur Verfügung gestellt. Hierdurch wird Forschung zur Vorbeugung, Erkennung und Behandlung von Tumoren unterstützt und verbessert. Die in die CCCF-Biobank eingelagerten Gewebeproben entstammen diagnostischen oder therapeutischen Maßnahmen von Routineuntersuchungen. Es handelt sich um überschüssiges Untersuchungsmaterial, das als vorhandenes Restmaterial vernichtet würde und bedeutet weder eine Veränderung für den Eingriff, noch wird es für die weitere Therapie oder Diagnostik benötigt. Ein daraus resultierendes gesundheitliches Risiko für die Patientin/den Patienten besteht zu keinem Zeitpunkt.

Aktuell können noch nicht alle zukünftigen medizinischen Forschungsziele und -möglichkeiten beschrieben werden. Diese können sich sowohl auf bestimmte Krankheitsgebiete (z.B. Krebsleiden, Herz-Kreislauf-Erkrankungen, Erkrankungen des Gehirns) als auch auf heute zum Teil noch unbekannte Krankheiten und genetische Defekte beziehen. Aus diesem Grund können an Biomaterialien möglicherweise auch genetische Untersuchungen durchgeführt werden, unter Umständen also Untersuchungen der Erbsubstanz und des gesamten Genoms.

Gewebeproben und Daten werden in der CCCF-Biobank unter standardisierten Qualitäts- und Sicherheitsbedingungen unbefristet aufbewahrt und nach dem Stand der Technik vor unbefugtem Zugriff gesichert. Gewebeproben und Daten können an Mitglieder der Universität Freiburg, anderer Universitäten, Forschungsinstitute und forschender Unternehmen (jeweils auch im Ausland) weitergeben werden. Hierbei  werden ausschließlich medizinische Forschungsprojekte unterstützt.

Alle Forscher*innen verpflichten sich, die geltenden Datenschutzrichtlinien einzuhalten.  Der Projektantrag in Kooperation mit dem Institut für Klinische Pathologie findet sich im Internet auf den Seiten der Biobank. Vorab müssen Forschungsprojekte von einer unabhängigen Ethik-Kommission unter ethischen und rechtlichen Aspekten geprüft und positiv bewertet worden sein. Für die Bereitstellung der Proben und Daten kann die CCCF-Biobank von den Nutzer*innen eine Aufwandsentschädigung erheben. Persönlich entsteht für Patient*innen kein unmittelbarer Vorteil oder Nutzen aus der Spende der Proben und Daten. Eine Information an Spender`*Innen über Untersuchungs- oder Forschungsergebnisse durch die CCCF-Biobank  erfolgt nicht.

Zu 5. Berichte über die Aktivitäten der Biobank  

Die CCCF-Biobank unterstützt klinische Studien, Drittmittelanträge sowie lokale, nationale und internationale Forschungsprojekte. Die Biobank berichtet turnusgemäß der AG Biobank und Genomics, sowie dem CCCF Board Shared Ressources in zweimonatlichen Abständen. Die Forschungsaktivität kann an der ausgewiesenen Publikationsliste nachvollzogen werden, sowie an den regelmäßigen Kongressteilnahmen, z.B. beim nationalen Biobankensymposium, Treffen des CCC-Netzwerkes und der Deutschen Gesellschaft für Pathologie (DGP).  

Zu 6. Maßnahmen der Qualitätssicherung  

Die CCCF-Biobank ist zertifiziert nach DIN EN ISO 9001:2015  seit dem 16.07.2019.  

Tätigkeiten der Biobank

Jede Probe wird vor Ausgabe einer Ausgangskontrolle unterzogen. Alle Prozesse von der Entnahme, der Einlagerung bis zur Ausgabe der Gewebeproben sind klar nachvollziehbar geregelt und mit dem Qualitätsmanagement der Routinelabore des Instituts für Klinische Pathologie verknüpft.  Die Biobank des Comprehensive Cancer Centers Freiburg verwaltet sowohl Kryo (-80° Celsius) als auch hiervon abgeleitete Derivate wie z. B. Paraffingewebe. Alle Projekte werden durch mindestens eine(n) erfahrene(n) Pathologin/Pathologen von Beginn bis Abschluss begleitet.

Weitere Serviceleistungen können auf Antrag und nach Absprache von allen CCCF-Mitgliedern genutzt werden.

Gewebebasierte Analysemethoden werden u. a. durch die Core Facility Genomics, die Proteomik Plattform und die Core Faciliy für Histopathologie und Digitale Pathologie zur Verfügung gestellt.

Die CCCF-Biobank beteiligt sich darüber hinaus an der Entwicklung neuer Methoden.
