The Department of Nephrology and General Medicine at the University Medical Center Freiburg, AG Prof. Dr. med. Carsten Bergmann, offers a PhD student position for a research project that aims for a better understanding of cystic kidney diseases and related ciliopathies. The project is part of the collaborative research center on nephrogenetics (SFB 1453) in Freiburg, Germany.
The research group focuses on the genomic identification and characterization of novel genes for cystic kidney disease and related ciliopathies. We overlook one of the worldwide greatest patient cohorts and regularly publish our data in top-ranked journals. New genes identified by different Next-Generation Sequencing (NGS) methods (WES, WGS, RNAseq) will be further investigated by a wide range of different cellular and molecular biological methods as well as in the zebrafish animal model. The project aims to better understand the complex protein network associated with the disease. Further characterization of newly identified proteins will add to a better understanding and provide new aspects for future therapeutic approaches.

Job description:
- analyses of human patient cells, knock-out/in cells (immunostaining, co-immunoprecipitation, luciferase reporter assays, antibody staining)
- the use of different cloning strategies
- validation and further characterization of new interaction partners from mass spectrometry analyses
- knock-down and knock-out analyses in the zebrafish animal model
- characterization of new mutant fish lines
- whole mount in situ hybridization and antibody staining in zebrafish
- signaling pathway analyses in cells and zebrafish (RNAseq, bioformatics)
- high resolution imaging (Confocal microscopy, Slide scanner, Expansion Microscopy)
- close collaboration with other members of our research group that enable insight into various other research areas (e. g. bioinformatics, clinical and molecular genetics)
What we are looking for:
- team-player with a positive and open mindset
- completed studies in the field of biology, medicine or related natural sciences
- high self-motivation and willingness to work independently
- diligent, accurate way of working
- willingness to work on the zebrafish animal model and to participate in routine work in the zebrafish facility
- previous knowledge in molecular biology, cell biology and microscopy desirable: molecular biology, cell biology, bioinformatics, statistics and / or microscopy
- previous experience in working with zebrafish is an advantage
