Hartmann lab: Mechanisms of leukemic infiltration

Research interest

Leukemia and lymphoma cell interactions with the microenvironment of bone marrow and secondary lymphoid organs are the main focus of our research group. We are interested in how chemokine signals and CD44 in acute myeloid leukemia (AML) and signals from the B cell receptor in chronic lymphocytic leukemia (CLL) converge with integrins to be integrated in downstream migratory and adhesive cues. We elucidate how this machinery is regulated by quiescence or activation of leukemic cells and which components are involved in homing, retention, and proliferation. Our previous data suggest that “adhesive spots” at the leukemia cell-stroma interface may function as signaling hubs. Current projects study the impact of downstream integrin adaptors and cytoskeletal integration on survival pathways in AML and CLL, with a particular focus on VLA-4 inside-out and outside-in activation and clustering induced by different signals from the hematopoietic microenvironment. We use time-lapse and real-time microscopy to study motility and adhesion under shear flow and static conditions, and cytometric tools combined with immunofluorescence to characterize changes in VLA-4 ligand affinity and cellular distribution.
